细菌蛋白质组双向电泳中“串联珠”现象初探

目的初步探讨细菌样品双向电泳中'串联珠'状蛋白点产生的原因。方法通过对比不同样品制备方法对双向电泳胶图的影响,分析'串联珠'状蛋白点出现的条件,并取'串联珠'状蛋白点进行胶内酶解及MALDI-TOF/TOF分析,寻找'串联珠'状蛋白点可能产生的原因。结果以含0.5%SDS的裂解液煮沸菌体的方法制备样品,就会产生'串联珠'状蛋白点。蛋白鉴定结果显示'串联珠'状蛋白点为同一蛋白,MALDI-TOF/TOF一级质谱峰没有出现明显的偏移峰。结论 '串联珠'产生的原因是由于SDS在加热条件下会对蛋白产生修饰或紧密结合作用,特别是相对分子质量较大的酸性蛋白。

新型铕(III)-谷氨酸-苯并咪唑三元配合物的合成及其电化学性质

以EuCl3.6H2O,谷氨酸(Glu)和苯并咪唑(BIM)为原料,在乙醇介质中于60℃反应6 h合成了新型的三元稀土配合物(1),其结构经UV-Vis,IR,TG-DTA,摩尔电导,化学分析和元素分析表征,确定1的化学组成为[Eu(Ⅲ)(Glu)3(BIM)]Cl3.3H2O。通过循环伏安法测定了1和EuCl3.6H2O在AcOH-AcONa缓冲溶液(pH 6)中的电化学性能,结果表明,在-0.4 V~-0.9 V,1和EuCl3.6H2O均表现出电化学活性,1的电化学活化中心是Eu3+,为准可逆体系,且还原峰电流与浓度和扫描速率的平方根呈良好的线性关系。
